Background
Neuronal Pentraxin 2 (NPTX2), also known as Neuronal Activity-Regulated Pentraxin (NARP), is a secreted protein predominantly expressed in neurons. It belongs to the pentraxin family and is rapidly induced by synaptic activity. Its primary function is at excitatory synapses, where it organizes the surface expression of AMPA-type glutamate receptors. By clustering these receptors, NPTX2 is crucial for strengthening synaptic connections and is a key mediator of homeostatic and experience-dependent plasticity. Dysregulation of NPTX2 is increasingly associated with various neurological disorders, including cognitive impairment, Alzheimer's disease, and schizophrenia.
